چکیده
The analysis of simple sequence repeats (SSRs) and bulk ground samples by droplet digital polymerase chain reaction (ddPCR) was investigated as an alternative to individual kernel testing for assessing the presence local cultivars in common wheat (Triticum aestivum L.) varietal blends. recent Protected Geographical Indication (PGI) ‘Pan Galego’ (Galician bread) requires that flour comprise a minimum 25% cultivars. As test compliance with this level, flours were prepared mixing commercial 0%, 5%, 20%, 30% ‘Caaveiro’ 100% used controls. A second performed set 5% ‘Caaveiro’. These mixed two different assess potential ability five SSRs identify percentage ‘Caaveiro’, constituting first reference use traceability specific autochthonous ddPCR using QX200 system platform targeted proportions across simulated range out SSRs, indicating they can be breads.

Digital polymerase chain reaction (dPCR) end point platforms directly estimate the number of DNA target copies per reaction partition, λ, where the partitions are fixed-location chambers (cdPCR) or aqueous droplets floating in oil (ddPCR). For use in the certification of target concentration in primary calibrant certified reference materials (CRMs), both λ and the partition volume, V, must be m...
متن کاملDevelopment of New Modified Simple Polymerase Chain Reaction and Real-time Polymerase Chain Reaction for the Identification of Iranian Brucella abortus Strains
Brucellosis is primarily a worldwide zoonotic disease caused by Brucella species. The genus Brucella contains highly infectious species that are classified as biological threat agents. In this regard, the identification of Brucella can be a time-consuming and labor-intensive process posing a real risk of laboratory-acquired infection to the laboratory staff. This stud...
متن کاملDetection of epidermal growth factor receptor mutation in lung cancer by droplet digital polymerase chain reaction
BACKGROUND Two types of epidermal growth factor receptor (EGFR) mutations in exon 19 and exon 21 (ex19del and L858R) are prevalent in lung cancer patients and sensitive to targeted EGFR inhibition. A resistance mutation in exon 20 (T790M) has been found to accompany drug treatment when patients relapse. These three mutations are valuable companion diagnostic biomarkers for guiding personalized ...
متن کاملSimple Sequence Repeats Amplification: a Tool to Survey the Genetic Background of Olive Oils
A reliable DNA extraction method for use on extra virgin olive oil based on a commercial kit was defined, and the possibility of using this DNA for fingerprinting the original cultivar was demonstrated. The genetic traceability of single-cultivar virgin olive oil from two cultivars (Carolea and Frantoio) was achieved by identifying the varieties from which they were produced. This involved the ...

Invasive aspergillosis (1 is a life-threatening condition in immunocompromised patients. An early diagnosis is of great importance because early treatment may resolve this potentially fatal infection. Recently, the polymerase chain reaction (PCR) has been used successfully in detecting specific DNA of several pathogen. In this study, nested PCR was used to detect DNA specific for A!.pergiflus s...
